纳污坑塘对古菌群落特征的影响

摘要

选取了3种(8个)不同污染程度的纳污坑塘,采用高通量测序技术对水体和沉积物中的古菌群落进行分析.结果表明,不同污染程度的纳污坑塘水体和沉积物中的古菌群落的组成、多样性和结构存在显著差异.在纳污坑塘水体中,影响古菌群落的主要环境因子为pH值(19.3%)和NH3-N(11.5%),群落的组装过程以确定性过程为主.轻度污染的指示类群为奥丁古菌纲、Altiarchaeaceae和甲烷八叠球菌属;中度污染的为甲烷微菌目和甲烷螺菌属;重度污染的为热球菌纲、甲烷硬脂菌科和亚硝化杆菌科.在纳污坑塘沉积物中,驱动古菌群落变化的环境因子主要包括pH值(7.9%)、Zn(7.3%)、Pb(7.3%)、Ni(7.1%)、Cu(6.8%)和Cr(6.3%).沿污染梯度,古菌群落的组装逐渐由随机性转为确定性过程,甲烷杆菌科、亚硝化球菌纲、深古菌纲、热原菌纲和Methanomethylophilaceae是轻度污染的指示类群;微古菌目、洛基古菌纲分别是中度和重度污染的主要指示类群.

Abstract

Three types (8sites) of sewage ponds with different pollution levels were selected, and high-throughput sequencing technology was used to analyze the archaeal communities in the water and sediments. The results showed significant differences in the composition, diversity, and structure of archaeal communities within the water and sediments of ponds across the different pollution levels. In the pond water, the main environmental factors influencing the archaeal community were pH (19.3%) and NH3-N (11.5%), and community assembly was primarily governed by deterministic processes. Indicator taxa for lightly polluted water were Odinarchaeia, Altiarchaeaceae, and Methanosarcina. Moderately polluted water was characterized by Methanomicrobiales and Methanospirillum. Heavily polluted water was indicated by Thermococci, Methanohalobiaceae, and Nitrosopumilaceae. In the pond sediments, key environmental drivers of archaeal community variation included pH (7.9%), Zn (7.3%), Pb (7.3%), Ni (7.1%), Cu (6.8%), and Cr (6.3%). Along the pollution gradient, the assembly process of archaeal communities gradually shifted from stochastic to deterministic. The indicator taxa for lightly polluted sediments were Methanobacteriaceae, Nitrososphaeria, Bathyarchaeia, Thermoplasmata, and Methanomethylophilaceae. Micrarchaeales was the main indicator for moderately polluted sediments, while Lokiarchaeia was the primary indicator for heavily polluted sediments.
