Interaction between benz[a]anthracene and pulmonary surfactant mixed phospholipid monolayers

Abstract:To investigate the effect of polycyclic aromatic hydrocarbons on pulmonary surfactant (PS) monolayers, mixed phospholipids of 1,2-dipalmitoyl-sn-glycerin-3-phosphate (DPPC) and 1-palmitoyl-2-oleoyl-sn-glycerine-3-phosphatidylcholine (POPC) were selected as PS models to study the interface chemistry reaction between benz[a]anthracene and DPPC/POPC mixed phospholipids. The results showed that benz[a]anthracene can significantly affect the phase transition of DPPC/POPC mixed phospholipid monolayers and weaken the stability of the monolayers. Furthermore, benz[a]anthracene disturbed the dense and ordered arrangement of phospholipid molecules. With the increase of the proportion of POPC, the adverse effects were aggravated. DPPC/POPC mixed micelles can enclose benz[a]anthracene promoting the dissolution of benz[a]anthracene.When the concentration of DPPC is 200mg/L, the apparent solubility of benz[a]anthracene is nearly three times that of aqueous solution and the solubilization was more obvious with the increase of POPC, which was mainly due to the fact that POPC was beneficial to the dispersion of micelles. We can draw a conclusion that benz[a]anthracene can inhibit the physiological function of PS, and PS can in turn affect the dissolution and migration of benz[a]anthracene, further increase the exposure risk of benz[a]anthracene. At the same time, the change of PS components caused by physiological conditions may lead to the difference of potential toxicity of benz[a]anthracene, and the lung injury of people with related lung diseases may be more serious.
